The easiest way to describe the difference is with an example:

A Trekkie shows up to a star trek convention having taped vulcan ears to his head, and thinks he looks cool.

A Trekker shows up to a star trek convention in full Klingon makeup, including facial prosthesis, full Klingon battle armor, a Bat'leth hanging off his back by a leather strap, 4 or 5 knives hidden on his armor, and can probably have a conversation in Klingon (either Standard Klingon or Clipped Klingon) with the other Trekkers there.
